Trincomalee begon ooit als een nederzetting van Indiase Tamils, die in de 19e eeuw naar Sri Lanka werden gehaald om te werken op plantages van de kolonisten. De vroegere Tamil-nederzetting was één van de oudste nederzettingen van het land. Tijdens de koloniale tijd hoorde Trincomalee bij Nederlands-, Portugees- en Brits-Sri Lanka, en is ook nog één jaar Deens geweest. De haven van Trincomalee was een belangrijk strategisch punt voor de handel van deze Europese kolonisten.
